布地奈德对哮喘大鼠肺部炎症及血清白细胞介素-5水平的影响 被引量:16

Influence of Budesonide Suspension on Pneumonic Anaphylacticitis and Level of Serum Interleukin-5 of(Asthma)-tic Rats

摘要 目的比较布地奈德雾化液与地塞米松对哮喘大鼠肺部过敏性炎症抑制作用及其对血清IL-5的影响,探讨布地奈德治疗哮喘与全身用糖皮质激素的差异。方法健康Wistar大鼠26只随机分为正常组、哮喘组、布地奈德组和地塞米松组。以卵清蛋白致敏激发法制作哮喘大鼠模型。布地奈德组每次激发前予布地奈德雾化吸入,地塞米松组每次激发予地塞米松(0.2mg/kg)腹腔注射。用ELISA试剂盒测定大鼠血IL-5水平。采血后取肺,作病理切片,HE染色观察。结果哮喘组大鼠肺及支气管的病理改变与其他各组比较有显著差异,布地奈德组与地塞米松组无显著差异,但两组与正常组比较有差异。哮喘组血清IL-5水平明显升高,与正常组比较有显著性差异(P<0.01)。布地奈德组及地塞米松组血清IL-5水平均下降,与正常组相比无显著性差异(P>0.05),与哮喘组相比均有显著性差异(P<0.01)。结论布地奈德局部吸入治疗可明显抑制哮喘大鼠肺部过敏性炎症反应,改善过敏性哮喘症状。 Objective To compare the inhibition effect of budesonide suspension and dexamethasone on the rats' pneumonic anaphylactieitis and serum IL - 5, and study the effect mechanism in treatment of asthma with glucocorticoids on systemic administration and with budesonide suspension. Methods Twenty - six healthy wistar rats were randomly divided into normal group, asthmatic group, budesonide suspension group and dexamethasone group. Asthma models were sensitized with egg albumin, Before sensitized, budesonide suspension group were administered inhalation of budesonide suspension, and dexamethasone group were administered abdominal injection of dexamethasone. The level of IL - 5 was measured with enzyme linked immunosorbent assay (ELISA) reagent box, lung was eviscerated after collecting blood, pathological slice was made. Results The rats' pathological changes of lung and bronchus were significant be twecn asthma group and other groups, while there were no significant difference between budesonide suspension group and dexamethasone group, but both of two groups showed slight difference with normal group. Compared with normal group, the level of serum IL - 5 in asthma group raised obviously, and there was significant difference (P〈0.05). Compared with asthma group, the level of serum IL-5 in budesonide and dexamethasone groups were lower, and there were significant differences (P 〈 0.05 in budesonide group, P 〈 0.01 in dexamethasone group); Compared with dexamethasone group and normal group, there were no significant difference from budesonide group(P 〉 0.05). Conclusion The treatment of inhalation of budesonide suspension can inhibit rats' pnetumonic anaphylacticitis, ameliorate rat anaphylactic asthma symptom.
