下肢缺血再灌注脂质过氧化损伤及维生素C的保护作用 被引量:2

Lipid peroxidation injury of lower limb after reperfusion and the protective effect of vitamin C

摘要 通过建立动物下肢缺血再灌注模型,检测血浆脂质过氧化物(MDA)和维生素E(VitE);观察下肢缺血再灌注脂质过氧化损伤严重性及损伤的恢复时期;应用大剂量维生素C(VitC)预防及治疗脂质过氧化损伤。结果发现:缺血4小时后再灌注,无脂质过氧化损伤发生:脂质过氧化损伤发生于缺血6小时血管再通的组织。观察缺血10小时、24小时后血管复通的实验组,发现缺血时间越长,再灌注后脂质过氧化反应增强越明显。缺血6小时后,干复灌注同时应用大剂量Vit C,发现再灌注后脂质过氧化反应无增强;缺血24小时后于复灌注时应用VitC,脂质过氧化反应减弱;表明大剂量Vit C能抑制或减轻下肢脂质过氧化损伤。 A model of canine hind limb is- chemia and reperfusion was established to in- vestigate the alterations and lipid peroxidation by measuring malondialdehyde MDA, or su- peroxide dismutase SOD. When reperfusion was pevformed 4 h after ischemia, MDA and SOD levels exhibited any significant change and no reperfusion injury was found. When reperfusion was done 6h after ischemia, MDA and SOD levels were found to change com- pared with those before ischemia P<0.05, indicating development of lipid peroxidation injury. Observation on 10h and 24h ischemia showed that the longer the ischemia, the high- er the MDA level. The degree of lipid peroxi- dation after reperfusion were correlated with ischemic time. After 6h of ischemia, when a large dose of Vit Cwas given at the time of reperfusion, MDA, SOD and Vit E levels did not change significantly. This suggested that lipid peroxidation did no develop. After 24h of ischemia and use of a large dose of Vit C at perfusion, MDA level increase was less than that when Vit C was not used P<0.05. It is conchuded that large doses of Vit C can inhibit or ameliorate lipid peroxidation.
