摘要
目的:观察褪黑素(MLT)对荷瘤小鼠的免疫调节及抗肿瘤作用。方法:以H22肝癌小鼠为模型,采用流式细胞仪技术及四甲基偶氮唑盐(MTT)比色法测定荷瘤小鼠的各项免疫指标。结果:MLT能提高荷瘤小鼠CD4+/CD8+值,协同IL-2提高外周血淋巴细胞及嗜酸性粒细胞数量,增强脾细胞NK及LAK活性,促进IL-2的诱生,并且MLT能抑制荷瘤小鼠的肿瘤生长,延长其存活时间,与IL-2存在明显协同性。MLT在体外对H22细胞无直接影响。结论:MLT可作为一种生物反应调节剂(BRM)在肿瘤的免疫治疗中发挥作用。
Objective:To observe the immunomodulation and antitumor effects of melatonin (MLT )in tumor-bearing mice. Methods:By the method of flow cytometry and MTT colorimetry,the immunological indexes of the H22 hepatoma-bearing mice were investigated. Results: MLT could increase the CD4+/CD8+cell ratio in the tumor-bearing mouse peripheral blood, cooperate IL-2 to promote the proliferation of lymphocytes and eosinophiles,increase the NK and LAK activity of spleenocytes, and enhance the production of IL-2 induced from the spleenocytes.MLT could also inhibit the tumor growth.prolong the life-span of the tumor-bearing mice in vivo and there was a synergetic action between IL-2 and MLT,while there was no effect on H22 hepatoma cells when MLT was used in vitro.Conclusion:MLT may have great effects on tumor immunotherapy as one of biological reation modulators(BRM).
