摘要
目的探讨肾上腺髓质素(adrenomedullin,AM)对心肌梗死大鼠心肌重构的影响作用。方法采用结扎左冠状动脉前支制作大鼠心肌梗死模型,并将术后存活的40只大鼠随机分为(1)心肌梗死对照组,(2)0.05nmol.kg-1.d-1AM组,(3)0.1nmol.kg-1.d-1AM组,(4)1.0nmol.kg-1.d-1AM组;另设假手术组。各处理组每日皮下注射AM。4周后测量各组大鼠心脏重量指数(hear weighy index,HWI),左室重量指数(left ventricular weight index,LVWI),左室非梗死区羟脯氨酸含量。结果与假手术组比较,心肌梗死组HWI、LVWI,羟脯氨酸均明显增加(P<0.01);与心肌梗死组比较,不同剂量AM组上述指标均显著下降,并呈剂量依赖性(P<0.01)。结论AM可有效改善心肌梗死大鼠心肌重构现象。
Objective To investigate the effects of adrenomedullin (AM)on myocardial remodeling after myocardial infarction in rats. Methods Left anterior descending coronary arteries were ligated to produce myocardial infarciton in rats, 40 rats which survived after coronary arteries ligation were randomly diveded into 4 groups: myocardial infarction group, AM (0.05nmol · kg^-1· d^ - 1 ) group, AM (0. 1 nmol· kg^-1· d ^-1 ) group and AM ( 1.0nmol· kg^-1· d^ -1 ) ; Sham-operate rats were selected as non-infarcted controis, subcutaneous injection of AM was given to the 4 groups everyday.4 weeks later, the parameters of heart weight index(HWI), left ventricular weight index(LVWI) and hydroproline concentration in left ventricular non-infarcted zone were measured. Results HWI, LVWI and hydroproline concentration increased significantly in myocardial infarction group as compared with those in shamoperated group( P 〈 0.01),and they decreased greatly in AM groups as compared with those in myocardial infarction group, and the decrease degrees were related to the dosage level of AM in different gropus. Conclusion AM can improve myocardial remodeling after myocardial infarction in rats.
