复方积雪草对UUO小鼠肾组织细胞转分化与肾纤维化的影响 被引量:9

摘要 [目的]研究复方积雪草对单侧输尿管梗阻后小鼠肾纤维化的防治作用。[方法]40只清洁级昆明种小鼠随机分为假手术组(A组)、单侧输尿管结扎组(B组)、复方积雪草治疗组(C组)、蒙诺对照组(D组)。于术后21天股动脉取血,检测血清肌酐、血白蛋白;留取梗阻侧肾脏制备病理切片,HE染色观察肾间质炎细胞浸润情况,Masson染色观察肾间质纤维化程度,并用RT-PCR技术检测肾组织α-SMA、VimentinmRNA表达。[结果]术后21d四组小鼠血清肌酐无显著性差异;与A组相比,B组肾组织α-SMA、VimentinmRNA表达增强;肾纤维化面积及程度加重;与B组相比,C组与D组α-SMA、VimentinmRNA表达明显减弱,肾纤维化程度减轻。[结论]复方积雪草能够下调肾组织α-SMA、VimentinmRNA表达水平,对肾纤维化有一定的防治作用。 [Objective]To evaluate the effect of Centella Asiatiea Compound on the process of renal fibrosis after unilateral ureteral obstruction in mice. [Methods] 40 male mice were randomly assigned to Sham operation group(A group), UUO group(B group), an Operation plus Centella Asiatica Compound group(C group), an Operation plus Fosmopril group (D group), 21 days later, blood sample was collected from each mouse to measure the level of creatinine and albumin. And the kidneys of the mice were sliced to make pathological sections to evaluate the results of the renal pathology. The mRNA expression of α-SMA and Vimentin was examined by reverse transcription-polymerase chain reaction (RT-PCR). [Results] There is no statistical significance among the lev els of creatinine in the four groups, After operation, the operated mice showed renal fibrosis. In comparison with A group, the mRNA expression of α-SMA and Vimentin in the interstitum of B group significantly increased (P≤0.01); In comparison with B group, Centella Asiatica Compound significantly decreased the mRNA expression of α-SMA and Vimentin in the interstitum,and it also improved the histological changes of the UUO renal tissue . [Conclusions] Centella Asiatica Compound can downregulate the mRNA expression of α-SMA and Vimentin in UUO mice. This demonstrates its potential ability to inhibit renal intersititium fibrosis caused by UUO in mice.
