摘要
目的:探讨糖尿病大鼠早期大血管病变中核因子-κB(NF-κB)及生长因子表达的变化及意义。方法:选用Wistar大鼠24只,随机分为正常对照组和糖尿病组,每组12只。以四氧嘧啶50mg/kg尾静脉注射制造糖尿病模型。10周后,免疫组化染色观察大鼠主动脉中NF-κB及血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)蛋白表达情况。结果:糖尿病大鼠主动脉表现为局限性内膜增厚,中膜浅层平滑肌细胞轻度增生、排列紊乱,病灶处内弹力板呈波浪状或交织状排列,有断裂分离现象。糖尿病组与对照组相比,主动脉NF-κB、VEGF及bFGF蛋白表达均增加,差异有统计学意义(均P<0.01)。结论:高血糖引起的NF-κB、VEGF及bFGF表达增加在糖尿病早期大血管病变的发生发展中可能有重要意义。
Objective: To study the significance of changes in expression of nuclear factor-kappaB (NF-κB)and growth factors in early diabetic macrovasculopathy. Methods: Twenty-four Wistar rats were randomly divided into two groups: normal control group and diabetic group. Diabetes was induced in experimental group by injection of alloxan 50 mg/kg via the tail vein. The expressions of NF-κB, vascular endothelial growth factor(VEGF) and basic fibroblast growth factor(bFGF) in aortas were studied by immunohistochemistry in 10 weeks. Results: The thickness of topical tunica intima of aortas increased in diabetic rats. In each focus, the elastic lamina existed asymmetric thickness and some had fissures and abruptions. The smooth muscle cells (SMCs) in superficial lamella showed slight proliferation and disorganized arrangemenL Compared with control group, the expressions of NF-κB, VEGF and bFGF in aortas significantly increased in diabetic rats (P 〈 0.01). Conclusion: The increased expression of NF-κB, VEGF and bFGF induced by hyperglycemia is of great significance in early diabetic macrovasculopathy.
