摘要 目的观察白细胞介素-1α(IL-1α)对体外培养的人眼小梁细胞(HTM)吞噬功能的影响。方法取人眼小梁组织进行小梁细胞体外培养,取传3~5代的人眼小梁细胞,加入不同浓度的IL-1α的DMEM-F12培养液,用倒置相差显微镜、光镜、电镜观察细胞吞噬功能的变化。结果小梁细胞吞噬乳胶微球数与IL-1α浓度(F=3.603,P=0.046)和作用时间(F=15.846,P=0.005)均有相关性(F=18.069,P=0.000)。结论IL-1α可以直接损害体外培养的人眼小梁细胞吞噬功能,其吞噬程度与IL-1α作用时间和浓度相关。 Objective To investigate the effect of interleukin-1α (IL-1α) on the phagocytic function in cultured human trabecular meshwork (HTM) cells. Methods HTM Cells were cultured and subcultured in vitro. The third to fifth passage of HTM cells were treated with different concentrations of IL-1α or vehicle in DMEM/F12 medium. 1000 of 0.85 μm microsphere/per cell were added to each well. The morphological changes of HTM cells were observed by an inverted phase-contrast microscope and electron microscope. The cellular phagocytic function of HTM cells was evaluated by counting the numbers of microsphere in 20 HTM cells using a light microscope. Results The break of HTM cell membrane was found under the electron microscope. The numbers of microsphere in HTM cells was significantly ( P = 0. 005 ) reduced in a dose-and incubation time-dependent manner( F = 3. 603 ,P = 0.046, F = 15. 846, P = 0. 005 ,respectively) . Conclusions IL-1α affects the phagocytic function of HTM cells that is correlated to the concentration and the incubation time.
