宫颈癌变进程中凋亡基因表达与HPV感染的相关性研究 被引量:6

摘要 目的探讨宫颈癌组织中凋亡相关基因Bcl-2、bax表达及宫颈上皮内瘤变组织中人乳头瘤病毒感染与Bcl-2、bax表达的关系。方法应用免疫组织化学方法对宫颈正常上皮、CIN和宫颈癌共183例进行Bcl-2和bax蛋白表达的检测;PCR检测正常上皮和CIN组织中HPV16、18型的存在。结果Bcl-2表达阳性率在从低级别CIN组到高级别CIN组逐渐增高,而bax的表达与之相反(P<0.05);CINⅡ、CINⅢ的Bcl-2的阳性表达率与HPV感染呈正相关;Bcl-2和bax在宫颈癌的表达率明显高于正常宫颈上皮(P<0.01);宫颈鳞癌Bcl-2表达与肿瘤分化程度呈正相关。结论HPV感染可能与诱导Bcl-2的过度表达,抑制细胞凋亡,促进组织增殖有关,从而在宫颈癌发生、发展中起重要作用。 Objective To study Expression of apoptosis regulating gene in cervical cancer and its Correlation with Human Papillomavirus Infection in Cervical intraepithelial neoplasia .Methods Immunohistochemical SP method was used to detect the expression of bcl 2/bax in 183 cases cervix uteri tissue , HPV16, 18 were detected with PCR. Results Expression rate of Bcl-2 and Bax showed significant difference among various degree of CIN(P〈0.05). Expression of Bcl-2 showed a positive correlation to HPV infection in CIN-Ⅱ, CINⅢ(P〈0. 05); The positive rate of Bcl-2 and bax expression was significantly higher in invasive squamous cell carcinoma than those in normol cervix uteri tissue (P〈0.01). There are positive correlation between Bcl 2 expression and tissue differentiation in invasive squamous cell carcinoma. Conclusion HPV infection inducing overexpression of Bcl-2 plays an important role in development and malignant chang of cervical carcinoma through suppressing Apoptosis and accelerating proliferation of cell.
