摘要
通过单因素比较和正交设计试验,研究了pH、温度、碳源、氮源、C/N等主要因素对木霉菌TD31菌株菌丝生长的影响。结果表明,pH、温度、碳源、氮源、C/N对木霉菌TD31菌株菌丝生长有明显影响。TD31菌株在pH2~11范围内均可生长,较适pH为2~7,最适pH为4;在10~35℃都可生长,25℃为最适生长温度;在无机碳源培养基上不能生长,可利用单糖、二糖和多糖,葡萄糖和果糖是最适碳源;在8种氮源中,以硫酸铵、硝酸铵、氯化铵和蛋白胨为氮源时,菌丝生长速率较高,尿素不是菌丝生长的适宜氮源。在C/N为10:1~60:1范围内均能较好生长,其中在C/N为50:1时,菌丝生长最快。TD31菌株菌丝生长的最佳培养条件是葡萄糖3%、(NH_4)_2SO_40.3%、pH= 4、25℃。
The hyphal growth of Trichoderma strain TD31 was affected by several factors, such as pH temperature, carbon source and C/N so on. The experiment was done by single -factor- comparison and perpendicular designing. The result showed that the effects were obvious : The pH suitable for growth was 2 - 11, the range for the better growth was 2 - 7, the optimum was 4 ; the suitable temperature was 10-35℃ and the optimum was 25℃. The hypha of Trichoderma strain TD31 could grow on the media with monosaccharide, disaccharides and polysaccharides and couldn' t grow on the medium with inorganic carbon source. The optimum carbon source was glucose and fructose. The growth speed was faster on the media with ammonium sulphate, ammonium nitrate,ammonium chloride and peptone as nitrogen source. Urine was not suitable for the hyphal growth. The hypha grew well on the media withC/N 10 : 1-60 : 1 ( The optimum ratio wasC/N50 : 1).
