摘要
根癌农杆菌介导的遗传转化法(ATMT)已广泛地应用于真菌的转化研究,该方法具有操作简单、转化效率高和转化子遗传稳定等特点。木霉作为土传植物病原菌的重要生防真菌,对其生防分子作用机制方面的研究也越来越受到关注,目前所面临的问题是如何基因突变和基因替换。ATMT法为木霉(Trichodermaspp.)的分子生物学研究提供了一个强有力的工具,在木霉的插入突变、遗传转化以及基因标记和克隆方面起着重要的作用。本文综述了根癌农杆菌介导木霉遗传转化的原理、特点、转化方法和应用,并且对有关发展趋势进行了展望。
Agrobacterium tumefaciens-mediated transformation (ATMT) has be widely applied to fungi due to its simplicity and high efficiency as well as stable transformants. Trichoderma spp. serves as an important fungal biocontrol agent to antagonize soil-borne plant pathogens and one problem with understanding the mechanisms underlying biological control in it is that the process of gene disruption and gene replacement. ATMT provides an efficient tool for the molecular genetic research on Trichoderma spp., which plays an important role in the fields of targeted or random insertional mutagenesis, genetic transformation, gene tag and cloning. Progress in transformation of Trichoderma spp. mediated by Agrobacterium tumefaciens and the application are summarized in this review .
