摘要
在大鼠肢体缺血—再灌注模型上观察脂质体携载前列腺素E1(PGE1)的保护作用。结果发现,脂质体携载PGE1治疗明显减轻缺血—再灌注肢体的肌肉水肿、减少中性粒细胞浸润,抑制肌肉组织及细胞线粒体的钙超载,抑制缺血—再灌注引起的血浆乳酸脱氢酶和组织蛋白酶D活性增加,降低血浆脂质过氧化物丙二醛含量和血浆内皮素含量。脂质体携载PGE1的治疗作用明显优于相同剂量的游离PGE1的作用。实验结果提示,脂质体携载PGE1对缺血—再灌注损伤具有潜在临床治疗价值。
The experiment was performed on the model of rat limb ischemia/reperfusion(I/R). It was found that treatment with liposome carried prostaglandin E 1 (PGE 1, 10 μg·kg -1 iv) significantly alleviate I/R induced muscular edema, attenuated neutrophil accumulation in the I/R tissue, inhibited calcium overload in the skelet muscle and cellular mitochondria, lowered activities of plasma lactate dehy drogenase and cathpsin D, and decreased plasma lipid peroxide formation and plasma endothelin content. The protective effects of liposome carried PGE 1 were obviously better than that of free PGE 1 at the same dose. The results suggested that liposome carried PGE 1 could have potential value for clinical therapy of I/R injury.
