摘要
目的初步研究转导人组织激肽释放酶基因对高血压大鼠的降压及其他作用。方法从质粒中扩增人组织激肽释放酶基因,定向亚克隆至pcDNA3.1载体,测序鉴定。Wistar大鼠,饮水中伺于10%果糖制备高血压模型。经静脉分别注入重组质粒及空白质粒。ELISA法检测大鼠血清激肽释放酶水平,尾套法测血压变化。常规测量大鼠胰岛素、血糖及甘油三酯。结果与对照组相比,大鼠血清中人组织激肽释放酶基因表达明显增加,并明显降低大鼠血压。结论转导人组织激肽释放酶基因使得高血压大鼠血压明显下降。
Objective To study the effects of human kallikrein gene delivery into hypertensive rats.Methods Human kallikrein gene was amplified from the plasmid and sub-cloned into plasmid pcDNA3.1 and the. recombinant plasmid was sequenced.Hypertension was induced by addition of 10% fructose to drinking water using Wistar rats. And then the recombinant plasmid and empty plasmid was delivered by intravenous injection. The level of the kallikrein in rat serum was assayed by ELISA, The systolic blood pressure was measured by the tail cuff method twice a week. Insulin, serum glucose and triglycefide were also detected. Results Compared with the control group, a single injection of human tissue kallikrein DNA caused reduction of systolic pressure clearly with the obviously increasing of kallikrein protein level, Conclusion Gene delivery of human tissue kallikrein could reduce hypertension in fructosefed rats.
