摘要
目的:研究罗通定在大鼠胆汁中的主要代谢产物。方法:灌胃给药后,收集大鼠胆汁,液液萃取,采用HPLC/MS/MS法分析鉴定罗通定的代谢产物。色谱柱Lichrospher-C18(5μm,4.6mm×250mm),流动相:甲醇-水-三乙胺(37630.063,V/V/V),冰醋酸调pH至6.3,检测波长281nm。结果:在大鼠胆汁中发现2个罗通定代谢产物,初步推测其结构为罗通定单羟基化后再与葡萄糖醛酸结合的产物。结论:罗通定在大鼠胆汁中主要以羟基化后的葡萄糖醛酸结合物形式排泄。
AIM: To investigate the main metabolites of mttmdine in rat bile. METHOD: Bile of rats after oral administration of rotundine (200 mg·kg^-1) was analysed by HPLC/MS/MS. Chromatographic analysis was performed on a Lichrospher-Cl8 column (5μm, 4.6mm×250 mm); mobile phase: methanol-water-triethylamine (37 : 63 : 0.063, V/V/V), adjusting pH to 6.3 with acetic acid. UV detector was set at 281 nm and positive ion mode was operated. RESULT: Two metabolites were found in rat bile, whose structure may be glucuronide conjugates of rotundine after demethylation. CONCLUSION: Conjugate with glucuronic acid after demethylation may be the main excretion pathway of rotundine in rat bile.
