层粘连蛋白的ELISA检测

Determination of serum laminin by ELISA

应用本室制备的两株抗人胎盘层粘连蛋白（ＬＮ）单克隆抗体３Ａ１５和１Ｂ５及ＬＮ标准，建立了测定血清ＬＮ的双单抗夹心ＥＬＩＳＡ法。本法稳定，检测下限为１０μｇ／Ｌ，在１０～５００μｇ／Ｌ之间呈线性关系。两份样本批内平均ＣＶ为６．８％和９．２％，批间ＣＶ为５．６％和６．１％，回收率平均为８８％。

Two clones of hybirdoma which secreted monoclonal atibodies named as 3A15 and 1B5 to laminin were developed. The 3A15 and 1B5 recognized the different antigenic epitope of laminin. A sandwich ELISA for determining serum LN concentration was established. The quantitative range is 10￣500 μg/L. For two serum samples, the average intra assay were 6.8% and 9.2% respectively, the average inter assay were 5.6% and 6.1% respectively, The average recovery rate was 88%. The normal range was found to be 20￣80 μg/L.
