摘要
通过基因转染技术将人MnSODcDNA载体导入中国仓鼠卵巢(CHO)细胞,建立了稳定表达有义和反义MnSOD的细胞系,并用此细胞模型初步探讨了MnSOD在电离辐射损伤中的作用,从正反两个方面说明了MnSOD对电离辐射敏感性的影响。
Manganese superoxide dismutase(MnSOD)is the most important antioxidant enzyme mainly lo-cated in mitochordria, MnSOD can protect cells from reactive oxygen species(ROS)injury by scanvengingOigenerated in the respiratory chain. Cell lines stably expressing human MnSOD cell lines were establishedby transfecting Chinese hamster ovary(CHO) ceIls with sense and antisense human MnSOD cDNA vec-tors. It is known that ionizing radiation could induce ROS,thus we investigated the role of MnSOD in ioniz-ing radiation injury and demonstrated the effect MnSOD on radiosensitivity. It is shown that overexpressionof MnSOD could protect and promotes the survival of CHO cells from ionizing radiation.
