摘要
使用动脉夹阻断基底动脉和两侧颈总动脉60min,然后恢复血供60min。在夹闭三血管前15min,腹腔注射绞股蓝总皂甙注射液,观察绞股蓝总皂甙对脑缺血再灌注损伤的保护作用。结果发现,绞股蓝总皂甙能显著减轻脑细胞内水、Na ̄+含量,K ̄+向细胞外移动减少,细胞内乳酸脱氢酶和肌酸激酶释放明显受抑,脑组织内乳酸积聚显著改善,丙二醛含量显著下降。表明绞股蓝总皂甙对大鼠全脑缺血再灌注损伤有明显保护作用。
Acute cerebral ischemia-reperfusion injury was produced in rat by clipping basilar artery and bilateral commoncarotid arteries for 60 minutes,and followed by removing the clips for another 60 minutes.Gypenosides in dosage of l00mg/kg body weight was injected intraperitoneally l5 minutes before clipping.Electroencephalography(EEG)wasmonitored,and cerebral water,Na+ and K+ content,lactate dehydrogenase(LDH)and creatine kinase(CK)activity,lactate and malondialdehyde(MDA)were measured to examine the protective mechanism of gypenosides on global cerebralischemia- reperfusion injury.The results showed that cerebral water and Na+ content were significantly lower ingypenosides group than in reperfusion group(P<0.01,P<0.01),lactate dehydrogenase and CK release from brainduring cerebral reperfusion were depressed significantly(P<0.01),(P<0.01),lactate and MDA level were falling ingypenoside(P<0.0l,P<0.01).These results indicate that gypenosides has a proctective effect on global cerebralischemia-reperfusion injury Its mechanism is related to the inhibition of brain edema,intracellular enzymes release,lactaeaccumulation and lipo-peroxidation.
