摘要 目的:研究经人白介素1受体拮抗剂(hIL-1ra)基因转染后的人颞颌关节髁突软骨细胞对IL-1作用的影响。方法:将hIL-1ra基因以阳离子脂质体介导转染体外培养的人颞颌关节髁突软骨细胞,在转染细胞和对照组细胞培养基中加入IL-1β,描记细胞生长曲线,计算细胞群体倍增时间,MTT法检测细胞增殖情况的变化,硝酸还原酶法测定培养液中NO2-/NO3-浓度的变化。结果:转染细胞与对照组细胞增殖情况存在差异,转染细胞高于对照组,硝酸还原酶法测定细胞培养液中NO2-/NO3-浓度,细胞培养液中NO的生成显著低于对照组(P<0.01)。结论:转染细胞hIL-1ra基因的表达对于IL-1作用存在拮抗和抑制作用。 [ Abstract] Objective:To investigate the effects of human i nterleukin-1 receptor antagonist (hlL-1 ra) gene trans- fection on condylar chondrocytes of temporomandibular joint (TMJ). Methods:Cultured condylar chondrocytes of TMJ were transfected by hlL-1 ra gene via cationic liposome. The proliferation of the transfected cells was examined by MTF assay. The nitrate reductase method was used to measure the nitric oxide (NO) synthesis in the supernatant of cell culture, normal TMJ chondrocytes without gene transfection were served as the controls. Results :The popula- tion doubling time(h) of transfected cells and the controls was 118.62 _+ 27.85 and 135.26 _+ 13.34 respectively. When the cells were exposed to IL-1 I3 the proliferation of transfected cells was increased more than that of the con- trois. NO concentration in transfected cells was lower than that in the controls with the presence or absence of IL-113 (P 〈 0.01 ). Conclusion:hlL-1 ra gene can antagonized the role of IL-1 β.
