通过重组DNA技术,从农抗5102产生菌10-22菌株中得到1株工程菌株WH-1,它的培养特征与出发菌株10-22有明显差别,前者气生菌丝白色,不吸水,基内菌丝浅黄色,可溶性色素淡黄;后者气生菌丝深灰,在大多数培养基均能吸水,基内菌丝暗柠檬色,可溶性色素淡褐。两者在28℃和37℃均可良好生长。菌株WH-1能产生菌株10-22所产生的3种5102抗生素,而且活性均更强,尤其是5102-Ⅰ几乎要高出10倍。菌株WH-1尚能产生菌株10-22不产生的1种活性物质(5102-Ⅳ)。该活性物质只能在琼脂培养基上产生。
A genetic engineering strain WH 1 has been derived from antibiotic 5102 producing strain 10 22 by means of recombinant DNA technology. It is distinctly different from the starting strain 10 22 in cultural characters. The former has white aerial mycelia that are non hygroscopic, pale yellow substrate mycelia and yellowish soluble pigment, while the latter has dark grey aerial mycelia that are hygroscopic in various media, dark citrine substrate mycelia and light brown soluble pigment. Both grew well at 28℃ and 37℃. Strain WH 1 is able to produce the three component of antibiotic 5102 produced by strain 10 22 and their activity is higher than that of strain 10 22, among them the activity of antibiotic 5102 Ⅰ produced by strain WH 1 is almost 10 times as high as that of strain 10 22. In addition, strain WH 1 can produce a component antibiotic 5102 Ⅳ, which is absent in strain 10 22, and grows only in agar medium. Its activity is very high in 24 hours after inoculation and reaches the peak in 48 hours, then it begins to decline. However, this active substance cannot be reproduced in liquid medium.
