摘要
目的观察阿司匹林(acetylsalicylic acid,ASA)对卵巢癌细胞SKOV3增殖和凋亡的影响。方法采用免疫组化法观察ASA对人卵巢癌细胞系SKOV3中COX-2基因蛋白和癌基因C-erbB-2的表达,流式细胞仪检测ASA对SKOV3细胞周期的影响,透射电镜观察ASA作用SKOV3细胞中凋亡情况。结果ASA可以明显降低癌基因C-erbB-2在SKOV3细胞中的表达,流式细胞仪结果显示:ASA对SKOV3细胞周期改变有影响,透射电镜发现在ASA作用下的SKOV3细胞有凋亡小体和坏死细胞形成。结论ASA对卵巢癌SKOV3细胞株具有抑制作用,能够对卵巢癌的防治发挥作用。
Objective The purpose of this study was to investigate the cellular effects of acetylsalicylic acid (ASA) on the growth of SKOV3 cell lines of human ovariancancer in vitro.Methods SKOV3 cell line was cultured with ASA. The cell cycle repartition was analysed by flow cytometry (FCM) ; The morphologic ehange and apoptosis of the cell were assessed under the electron microscope (EM). Immunohistochemical assessment was performed for alterations in expressions of COX-2 and C-erhB-2 enzymes. Results The expression of C-erbB-2 was decreased following treatment with ASA. The cell apoptosis and necrosis were detected with ASA through EM. The Cycle of cell was retention in G0/G1 by ASA (10^-2mol/L)treated. Conclusions Our suggests additional study for the use of ASA, as a stagy of ehemoprevention of ovarian cancer.
