胰岛素样生长因子-Ⅰ(Insulin-likegrowthfactor-Ⅰ,IGF-Ⅰ)是机体生长、发育和代谢的一个重要调控因子,同时也是生长激素(growthhormone,GH)启动生长活性的主要介导者。成熟的IGF-Ⅰ是由70个氨基酸残基组成的碱性单链多肽,因与胰岛素同源而得名。猪IGF-Ⅰ基因组DNA全长大于80kb,至少由6个外显子(exon1 ̄6)和5个内含子(intron1 ̄5)构成。虽然IGF-Ⅰ是一个单拷贝基因,但是,由于选择性拼接、不同的polyA位点以及潜在的多个启动子(promotors)的使用,使它以一种复杂的方式转录和处理而产生了多个成熟的mRNA拼接变异体,结构极为复杂。就猪IGF-Ⅰ基因的结构和功能等方面的最新研究进展进行综述。
Insulin-like growth factor-Ⅰ(IGF-Ⅰ ) is an important regulator of growth, development, metatabolism, and is the primary mediator of the growth-promoting activity of growth hormone (GH) in mammals. The mature IGF-Ⅰ is a 70-amino acid reside, basic single chain polypeptide which shows structural homology with insulin; it is expressed from a gene consisting of at least six exons (exonsl-6) separated by five introns(introns 1-5) and spanning more than 80 kb of genomic DNA. This single-copy gene is transcribed and processed in a complex manner to produce a large number of mature mRNA species as the result of alternative splicing, differential polyadenylation site usage, and, potentially, the use of multiple promoters. This article reviewed the latest research progress of structure and function of the pig IGF-Ⅰ gene.
