大鼠股神经感觉纤维在脊髓胶状质中的纵横定位

摘要 实验于2004-07/2005-12在咸宁学院解剖神经组化实验室完成。选用20只成年健康Wistar雄性大鼠,将其按随机抽签法分为2组:切断股神经组和切断神经根组,每组10只。切断股神经组;9只切断右侧股神经组,另1只仅切开股内侧皮肤,分离皮下组织,不切断股神经,以作对照。切断神经根组:9只切断右侧T13,L1,L2脊髓背根,另1只仅切开背部皮肤,分离肉至椎板不切断背根,以作对照。切取T12,13和L1~4数段脊髓,滑行切片。以Gomori硝酸铅法及Takeuchi鄄Tanove改良的硝酸铅法同时显示两组相邻切片的酸性磷酸酶活性,部分切片的孵育液内加0.02mmol/L氟化钠,单独显示抗氟化物酸性磷酸酶,以作对比。结果大鼠20只均进入结果分析。将胶状质由内向外划分均等于约10个(1~10)小区。切断神经根组:切断右侧T13和L1熏2脊髓背根后,L1节损伤侧整个胶状质酸性磷酸酶活性消失;T13和L2节的外侧份(5~10)区酶活性消失,内侧份(1~4区)保留。切断股神经组;切断右侧股神经后,酶活性消失按首尾方向主要分布在L1~3节。按水平方向酸性磷酸酶活性消失区,主要分布在损伤侧胶状质内侧1/2(2~5区),最内侧的一小部(1区)不消失。综合9只的情况看,其中L1节的酶活性消失区占2~4区,L2节占2~5区,L3节占3~6区,有越向尾侧越向朝外扩展的趋势。模拟手术对照动物伤侧胶状质酸性磷酸酶活性未见明显改变。说明感觉纤维在脊髓胶状质的定位投射柱有越向尾侧越朝外扩展的趋势。 The experiment was conducted in the Laboratory of Nerve-disSection and Histochemistry, Xianning College from July 2004 to December 2005. Twenty adult healthy Wistar male rats were selected and randomly divided into 2 groups: femoral nerve resection group and nerve root resection group with 10 rats in each group. Nine rats in the femoral nerve resection group were resected of the left femoral nerve, while the other rat was resected of the skin in interfeminium to isolate the subcutaneous tissue without resection of femoral nerve for control. Nine rats in the nerve root resection group were cut off the right spinal cord dorsal roots of T13, L1 and L2, while the other rat was cut off the skin on the back to isolate until the vertebral plate without resection of dorsal root for control. The spinal cord of T12,13 and L1-4 were cut off for slipping slices. Gomori lead nitrate method and Takeuchi-Tanove improved lead nitrate method were adopted to manifest the activity of tartrate-resistant acid phosphatase (TRAP) in adjacent slices of both groups, and 0.02 mmol/L of sodium fluoride was added into the incubation fluid of slices to singly show the TRAP activity of anti-fluoride, taken as the control. A total of 20 rats were involved in the analysis of results. The gelatinous substance was equally divided into 10 sections (Section1-10) in an ento-ectad manner. Nerve root resection group: Rats were cut off the right spinal cord dorsal root T13, L1 and L2, and the TRAP activity of total gelatinous substance in the affected side of L1 disappeared. The TRAP activity in external sections (5-10) of T13 and L2 disappeared, while that in the internal sections (1-4) were maintained. Femoral nerve resection group: The TRAP activity disappeared after resection of the right femoral nerve, which was mainly, distributed on L1-L3 segments in a head-tall direction. The TRAP activity horizontally disappeared, which was mainly distributed in 1/2 parts of the internal side in affected gelatinous substance (section 2-5), while it did not disappeared in the furthest part (section 1). Study on 9 rats showed that the TRAP activity of L1 disappeared in 2-4 sections, and that of L2 disappeared in 2-5 sections, and that of L3 disappeared in 3-6 sections, which were in an outward-extending trend as approaching to the caudal ward. There was no significant difference in TRAP activity in gelatinous substance of the affected side in control rats, which indicated that the located projecting column of sensory fiber on the gelatinous substance of spinal cord had an outward-extending trend as approaching to the caudal ward.
