摘要
目的探讨链脲佐菌素(streptozocin,STZ)致迟发型大鼠1型糖尿病模型建立的方法。方法将8只大鼠随机分为对照组3只,实验组5只;实验组将福氏完全佐剂(CFA)与小剂量STZ联合使用,连续3周大鼠腹腔注射,制造Wistar大鼠1型糖尿病模型,并对2组大鼠的进食量、饮水量、尿量、体重、血糖及尿糖进行密切监测,同时对实验组及对照组大鼠进行葡萄糖耐量实验(OGTT)。结果3只大鼠(60%)成模,实验组大鼠表现出多饮、多食、多尿、高血糖、高尿糖的1型糖尿病典型症状,与对照组大鼠的各项指标差异均有统计学意义(P<0.05),且实验组大鼠表现出葡萄糖耐量异常。结论CFA与小剂量STZ联合使用可制造Wistar大鼠1型糖尿病模型。
Objective To explore the method of establishing delayed type Ⅰ diabetic rat model induced by streptozocin (STZ). Methods Eight rats were randomly divided into two groups. Five rats in experimental group were induced by combined application of low-dose STZ with complete Freund's adjuvant (CFA), STZ with CFA was injected continously into rats' peritoneal cavity for three weeks. The changes of diet amount, drinking amount, urinary amount, body weight, blood sugar and urinary sugar were monitored. The OGTF were performed in the rats of both groups. Results 60% of rats could be successfully induced to typical diabetic animal model. As compared with the rats in control group, the rats in experimental group had typical type Ⅰ diabetic symptoms, such as the increase of diet, drinking, urine and body growth. There were significant differences in these parameters between the two groups (P 〈 0.05). The rats in experimental group showed obvious OGTF abnormality. Conclusion Rat model with type Ⅰ diabetes mellitus could be induced by injecting low-dose STZ with CFA into rats' peritoneal cavity.
