摘要
目的利用人脐静脉内皮细胞(HUVEC)构建人体外结直肠癌三维血管生成模型。方法原代培养HUVEC,通过Matrigel形成三维细胞培养系统,并在该系统内共培养HUVEC和结直肠癌LoVo细胞,观察HUVEC的血管生成。结果HUVEC和结直肠癌LoVo细胞成功共培养,形成了不连续的三维类毛细血管样结构。结论所构建的结直肠癌血管生成体外三维模型能为研究直结肠癌的血管生成过程提供了一个可供选择的体外模型,也能为直结肠癌血管生成干预和抗血管生成药物的筛选提供了一个更接近体内生理状态的研究平台。
Objective To develop a novel in vitro angiogenesis model of colorectal cancer with human umbilical vein endothelial cells (HUVEC). Methods A three-dimensional (3-D) cell culture system was formed by Matrigel,and HUVEC and colorectal cancer LoVo cell lines were co-cultured in this 3-D cell culture system. The angiogenesis of HUVEC in vitro was observed. Results HUVEC and LoVo cell line were successfully co-cultured. Capillary-like tubes were formed unconsecutively. Conclusion The 3-D angiogenesis model of colorectal cancer constructed in this study could be an alternative in vitro model for angiogenesis process of colorectal cancer, It mimicked the physiological state in vivo and could also be a research base for screening antiangiogenic drug.
