COC_1/DDP亚细胞中波形纤维蛋白的肽质谱指纹分析

摘要 目的寻找与鉴定卵巢癌耐药株中的差异表达蛋白质。方法应用高分辨二维凝胶电泳分离卵巢癌细胞中差异表达的波形纤维蛋白,并通过基质辅助激光解吸电离飞行时间质谱对其进行分析与鉴定。结果获得了分辨率和重复性均较佳的凝胶电泳图谱,图像分析显示波形纤维蛋白表达明显下调。结论与其他方法相比,应用凝胶电泳和生物质谱联用技术分析与鉴定未知蛋白具有简单快捷的特点。 Aim To search and identify the different expressed protein of subline COC1/DDP. Methods An obviously different expressed protein spot was separated by two dimensional electrophoresis, then analyzed and identified as vimentin using MALDI-TOF mass spectrometry. Results The good 2-DE images including resolution and reproducibility was obtained. The different protein expression analysis indicates that vimentin expression is obviously upregulated. Conclusion The results indica(e that the method has more advantage for identification unknown protein than the others by using 2-DE gel electrophoresis tenique combined with bio-mass spectrometry.
