摘要 目的建立胶滴肿瘤体外药敏检测技术,探讨其在肿瘤个体化治疗中应用的可行性。方法用人类肿瘤细胞系建立胶滴肿瘤体外药敏检测技术,并用该技术对50例不同肿瘤类型的原代肿瘤标本进行体外药敏检测实验研究,每种肿瘤标本分别进行了4~5种抗肿瘤药物的敏感性测定。结果在体外成功建立了胶滴肿瘤药敏检测技术,标本整体评价率为82.0%(41/50),检测结果与临床经验有效率间有较好的符合性。结论CD-DST体外肿瘤药敏检测技术是一种较好的药敏检测方法,标本可评价率为82.0%(41/50),所需标本量小,可反映病人对不同抗肿瘤药物敏感性的差异,检测结果与临床经验有效率有较好的符合性,对抗肿瘤药物的体外筛选和对个体化治疗具有实际应用价值。 Objective To establish Collagen gel droplet culture and explore its clinical feasibility of application. Methods drug - sensitivity test ( CD - DST) technique The human tumor cell lines were used to find out the best cuhure conditions of collagen gel droplet culture drug - sensitivity test ( CD - DST). The chemosensitivity of 4 or 5 anticancer drugs was assessed by the CD - DST in the ovarian and uterine cancer (n =18), gastrointestinal cancer (n=15), lymphoma (n =8), lung cancer (n=5), liver cancer (n= 4). Results We established the collagen gel droplet culture drug - sensitivity test ( CD - DST) by using the human tumor cell lines. The CD - DST can evaluate the efficacy of anticancer drugs in diverse kinds of tumor specimen. There was a statistically correlation between clinically reported response rates and the re- sponse rates obtained by CD- DST. The overall tumor evaluation rate was 82. 0% (41/50). Conclusion The CD - DST is a good chemosensitivity assay methods with the advantage of high overall predicting value and a comparatively small number of ceils, so that it can solve the issue of minor specimen source. Our re- sults in showed the in vitro sensitivity for each drug is a significant correlation with clinically reported re- sponse rates. It appears to be useful in exploring the possible combination plots and principles, evaluating the efficacy of existing chemotherapy, and optimize chemotherapy on an individual basis.
