摘要
以胶质芽孢杆菌(Bacillus mucilaginosus)(硅酸盐细菌)为材料,经液体发酵培养后,通过碱提醇沉法得到胞外多糖粗提物。紫外光谱分析表明,该粗提物不含有核酸和蛋白质,经DEAE-纤维素离子交换柱层析分离纯化得到多糖纯品。通过Sephadex G-100凝胶柱层析和醋酸纤维薄膜电泳鉴定,其为均一组分不含有其他多糖;化学法分析表明,多糖纯品含有氨基葡萄糖、葡萄糖醛酸,不含有硫酸根。
Crude polysaccharide was extracted separately from Bacillus mucilaginosus and purified by in-exchanging chromatography on DEAE-Cellulose columns. It was proved to be homogenous composition by ultraviolet spectrum, chromatography on Sephadex G-100 column and cellulose acetate membrane electrophoresis （ CAME）. The results also indicated that the polysaccharide was mainly made up of glucosamine and glucuronic acid identified by chemical methods.
