摘要
采用心脏停跳再复苏模型,以原位末端标记和形态学改变为指标,研究全脑缺血再灌注是否诱导神经细胞编程性死亡。结果显示,HE染色下,缺血再灌注后海马组织细胞体和核固缩,细胞周间隙增宽,无炎性细胞浸润。原位末端标记下,有散在的细胞核着色,可见半月状小块,表明有DNA裂解。证实脑缺血再灌注诱导神经细胞凋亡。提示神经细胞编程性死亡参与脑复苏中的损伤过程。
rogrammed cell death(or Apoptosis)is a major form of cell death,characterized morpho-logically by chromatin condensation and biochemically by endonuclease cleavage of DNA into o-lionucleosomal fragments.In this paper,we investigated whether brain ischemia-reperfusioncauses programmed neuronal death in dogs.The result showed characteristic features,such ascondensation of the chromatin,membrane blebbing by HE staining method and DNA degradationby a terminal deoxynucleotidyl-transferase(TdT)-mediated dUTP-biotin nick end-labelingmethod(TUNEL).These evidences support that brain ischemia-reperfusion induces programmedneuronal death.
