单纯疱疹病毒胸苷激酶基因表达对人肝癌细胞的生长抑制

摘要 使用转导有单纯疱疹Ⅰ型病毒胸苷激酶(HSV1-TK)基因的人肝癌SMMC-7721细胞株建立了裸鼠移植瘤模型,并进行了在体和体外试验性无环鸟苷(ACV)治疗。结果发现基因转导后的肝癌细胞表现出对ACV的敏感性,肝癌细胞生长抑制与ACV的剂量有关;用药组裸鼠除平均瘤重明显低于对照组外,还可见细胞核分裂指数降低,伴有细胞变性和坏死。结果表明:HSV1-TK基因经逆转录病毒转导后,用ACV诱导表达明显抑制了人肝癌细胞的生长。 We used the human hepatocellular carcinoma cell line(SMMC-7721) transduced with the herpes simplex virus I thymidine kinase(HSV1-tk)gene to establish a xenografted nude mice tumor model and used the transduced cells and tumor model to observe in vitro and in vivo the acyclovir(ACV) effect experimentally.The transduced cells rendered them sensitive to killing by ACV and the effect was related to doses of ACV. In addition to significant reductions in weight of residual tumors in ACV treated animals, the reduce of cell nuclear mitotic index and the changes of cellular degeneration and necrosis were noted. This study demonstrates that retroviral vector transfer of the HSV1-tk gene followed by ACV can significantly inhibit the growth of human hepatocellularcarcinoma cells.
