摘要
目的探索大鼠骨髓间充质干细胞(MSCs)同种异体移植后在受体大鼠模型肝内定居分布情况。方法利用密度梯度离心法分离、纯化培养大鼠骨髓间充质干细胞。取第三代MSCs经CFSE标记后从门静脉植入同种异体正常组和肝损伤组大鼠体内,于移植后的第14d取出的肝脏,通过冰冻切片行荧光检测,研究MSCs在大鼠肝脏内定居分布情况。结果经门静脉移植的MSCs在正常组和肝损伤组的肝脏内均能定居,且定居的细胞数量上肝损伤组明显多于正常组。结论经门静脉移植的MSCs可以定居于肝脏内,且定于肝脏的干细胞数量与肝脏是否损伤密切相关。
Objective To observe the distribution and amount of the mesenchymal stem cells(MSCs) after transplantation in rats. Methods BMSCs were isolated from femur in the rat by the density gradient centrifugation with Percoll solution and purified by the characteristic of the tissue culture plastic. MSCs of the third generation, after labeled by CFSE, were injected into portal vein of rats in normal group and hepatic lesion group . At the 14th day after transplantation, liver samples were collected. The distribution of MSCs in the frozen section was observed by fluorescence . Results MSCs could be found in all livers of rats. Furthermore, the quantity of MSCg in the column of damaged liver was obviously over that of normal liver. Conclusion MSCs transplanted through portal vein can successfully reside in the liver of rats and the homing status of MSCs is closely associated with quantity of stem cells and the damage status of liver .
