摘要
使用丙酮提取番茄果实中的色素,用反相高效液相色谱测定坚熟期番茄中的α-胡萝卜素、β-胡萝卜素和番茄红素。流动相梯度A为乙腈和水(97:3,v/v),流动相梯度B为乙酸乙酯,流动相B在80min内由0上升到100%,流速为0.5mL/min,500nm检测波长下进行检测,α-胡萝卜素、β-胡萝卜素和番茄红素的回收率依次为:90.87%~92.60%,90.22%~92.75%和91.08~93.48%。坚熟期番茄果实中检测到α-胡萝卜素、β-胡萝卜素和番茄红素的含量分别为3.718、78.48和34.92mg/kg。
A reversed -phase high performance liquid chromatographic method was used to determine carotenoids extracted with acetone from hard ripening stage tomato fruit. Mobile phase A consisted of acetonitrile and water (97:3,v/v) and mobile phase B was ethyl acetate. Binary gradient was performed linearly with the original of 100% A and finished in 80 minutes with 100% B. Carotenoids were detected at 500nm. It was shown that recoveries of α-carotene,β-carotene and lycopene were 90,87%-92.60%,90.22%-92.75% and 91.08%-93.48% o The hard ripening stage tomato contained α-carotene (3.718mg/kg), β-carotene (78.48mg/kg) and Iycooene(34.92mg/kg).
