摘要
目的:建立以毛细管电泳法测定人工牛黄中胆酸含量的方法。方法:运行缓冲液为30mmol/L十二烷基硫酸钠+40mmol/L磷酸氢二钠溶液(pH=9.0),进样量为50mbar×5s,分离电压为20kV,温度为30℃,检测波长为210nm。结果:胆酸检测浓度在60.6~970.0μg/ml范围内线性关系良好(r=0.9980),平均回收率为98%~102%(RSD<5%)。结论:该方法简捷、快速、准确、重现性好,可用于该药材的质量控制。
OBJECTIVE:To establish a capillary electrophoresis (CE) method for the determination of the cholalic acid in artificial bezoar, METHODS: The 30mmool/L sodium dodecyl sulfate plus 40 mmool/L dibasic ,sodium phosphate were used as buffer solution (pH = 9.0) with the sample size of 50mbar × 5s at 30℃ , the detection wavelength was 210nm and the separation voltage was 20kV. RESULTS: The linear range of cholalic acid was 60.6-970.0μg/ml (r = 0.9 980), the average recovery rate was 98%- 102% (RSD〈 5% ) ,CONCLUSION: The method was simple, rapid, accurate and reproducible for the quality control of artificial bezoar.
