细胞外三磷酸腺苷对大鼠脊髓损伤后微管相关蛋白-2表达及运动功能恢复的影响 被引量:1

Effect of extracellular ATP on the expression of MAP-2 and the recovery of motor function after spinal cord injury in rats

摘要 目的研究细胞外三磷酸腺苷(ATP)对大鼠脊髓损伤后微管相关蛋白-2(MAP- 2)表达运动功能恢复的影响。方法健康成年Wistar大鼠66只,随机取6只作为正常对照组.余60只制作成脊髓打击伤动物模型,随机分为两组:A组(ATP组)和B组(对照组),每组30只大鼠。伤后1,3,7,14,28 d取材,应用免疫组织化学方法观察MAP-2的表达,采用计算机图像分析系统,进行定量分析;并用改良的Tarlov评分观察大鼠脊髓损伤后运动功能的恢复情况。结果脊髓损伤后14 d和28 d,A组大鼠MAP-2的表达明显强于B组(P<0.05);损伤后14 d和28 d,A组大鼠改良Tarlov评分明显大于B组(P<0.05).结论细胞外ATP能促进大鼠损伤脊髓表达MAP-2,并能促进大鼠脊髓损伤后运动功能的恢复。 Objective To investigate the effects of extracellular ATP on the expression of ( Microtubule associated proteins MAP-2) and the recovery of motor function after spinal cord injury in rats. Methods Six rats were selected randomly from 66 adult healthy Wistar rats as the normal control group, the rest animals were divided into two groups after contusion injury was performed by drop weight method with Allen irapactor: Group A (ATP group) and Group B (control group), each group contained thirty rats. At days 1, 3, 7, 14, and 28 after injury, the rats were killed, the expression of MAP-2 was detected with immunohistochemistry. The expression of MAP-2 in the adjacent area was quantitatively analyzed with a computer image analysis system. The recovery of motor function after spinal cord injury was assessed with improved Tarlov scores, Results The expression of MAP-2 was higher in Group A than in Group B after spinal cord injury in rats. Significant difference was revealed by the expression of MAP- 2 between the two groups at days 14 and 28 after injury (P 〈0.05). Improved Tarlov scores in Group A were higher than in Group B after spinal cord injury in rats. Significant difference was revealed by improved Tarlov scores between the two groups at days 14 and 28 after injury ( P 〈 0.05). Conclusion Extracellular ATP can promote the expression of MAP-2 in spinal cord and the recovery of motor function after spinal cord injury in rats.
