趋化因子SDF-1α及其受体介导涎腺腺样囊性癌细胞的趋化与侵袭 被引量:1

摘要 探讨趋化因子SDF-1α及其受体CXCR4对涎腺腺样囊性癌细胞趋化与侵袭活性的促进作用。采用RT-PCR法检测涎腺腺样囊性癌肺高、低转移细胞株ACC-M和ACC-2中CXCR4 mRNA的表达;Boyden趋化小室法检测在SDF-1α作用下ACC-M细胞和ACC-2细胞的趋化与侵袭活性;检测CXCR4 mAb对ACC-M细胞和ACC-2细胞趋化活性和侵袭活性的抑制作用。结果显示:2株涎腺腺样囊性癌细胞中均存在不同程度CXCR4 mRNA的表达,其在肺高转移细胞株ACC-M中的表达显著高于肺低转移细胞株ACC-2;SDF-1α对2种细胞均具有趋化活性和侵袭活性,且对ACC-M细胞的趋化活性和侵袭活性显著强于ACC-2细胞;CXCR4 mAb能够抑制涎腺腺样囊性癌细胞的这种趋化活性和侵袭活性。趋化因子SDF-1α及其受体CXCR4能够介导涎腺腺样囊性癌细胞的趋化与侵袭。 The enhancement of the chemotactic and invasive activities to salivary adnoid cystic carcinoma cells mediated by chemokine SDF-1α and its receptor was investigated in the present study, in which the expression of CXCR4 mRNA was assayed in the salivary adnoid cystic carcinoma cell lines with high and low lung metastatic potentials, ACC-M and ACC-2 cell lines respetively by RT-PCR, and the the chemotactic and invasive activities of these cell lines induced by SDF-1α and its receptor CXCR4 were determined by means of Boyden chamber method. The inhibitory effect of anti-CXCR4 mAb on the chemotatic and invasive activities of these cell lines was also investigated. It was demonstrated that different levels of CXCR4 mRNA expressions could be detected in these two cell lines of salivary adenoid cystic carcinoma cells and the expression in ACC-M cells was significantly higher than that in ACC-2 cells. In addition, SDF-1α could enhance the chemotactic and invasive activities in these two cell lines, with higher activities in ACC-M cells in comaprison with those in ACC-2 cells. Both the chemostaxis and invasion in these cell lines could be blocked by treatment with anti-CXCR4 mAb. It is concluded that the chemostaxis and invasion of salivary adenoid cystic carcinoma cells can be faciliated by chemokine SDF-1α and its receptor CXCR4.
