Determination of baicalin content in suspension culture of Scutellaria baicalensis Georgi cells

目的 建立测定悬浮培养黄芩细胞中黄芩苷含量的方法。方法 采用HPIE法，色谱柱为Agilent Zorba×80A Extend-C18柱（150mm×4．6mm，3．5μm），流动相为甲醇-0．5％磷酸（50：50），柱温30℃，流速1．0ml·min^-1，检测波长280nm。结果 线性范围为0．244-2．440μg（r=0．9996），平均回收率为99．17％，RSD=1．39％（n=6）。结论 所用方法简便、准确、重复性好，可作为规模化生产黄芩药材细胞质量控制的有效方法。

OBJECTIVE To determine the baicalin content in suspension culture of Scutellaria baicalensis Georgi cells. METHODS An HPLC method was adopted. The separation was performed on Zorba × 80A Extend C18 column（ 150 mm×4.6 mm, 3.5 μm） with a mobile phase of methanol - 0.5 % phosphoric acid （50 ： 50 ）. The flow rate was 1.0 ml·min ^-1 and the detective wavelength was 280 nm. RESULTS The linearity of the method was well within the range of 0.244 to 2. 440 μg （ r = 0.9996） and the average recovery of baicalin was 99.17% with RSD of 1.39% （n =6）. CONCLUSION The method is simple, accurate and reliable. It can be used for quality control of Suctrllaria baicalensis cell production in factory in future.
