外周血CK19、MMP-9和MDR1水平评定体外药敏的价值 被引量:1

Measuring CK19,MMP-9 and MDR1 in Peripheral Blood to Evaluate the Value ofChemosensitivity on Breast Cancer in Vitro

摘要 目的检测乳腺癌患者外周血中细胞角蛋白19(CK19)、基质金属蛋白酶9(MMP-9)和多药耐药基因1(MDR1)的表达量,评定参照体外药敏选择化疗方案的价值。方法采用荧光定量RT-PCR技术,检测117例乳腺癌患者化疗前后外周血中CK19mRNA、MMP-9mRNA和MDR1mRNA的表达量。结果实验组较对照组外周血中CK19、MMP-9和MDR1mRNA的表达量ΔCt值变化有显著差异,平均差值分别为-5.2105/-3.9938、-5.3312/-2.6930和0.6750/3.6140。结论实验组较对照组,外周血中CK19、MMP-9mRNA的表达量显著下降,而对照组MDR1表达量的增高较实验组更显著,提示在现行的循证医学基础上增加体外药敏实验结果这一参考因素,有可能选择更敏感的化疗药物,提高个体化治疗的效果。 Objective The study aim to measure CK19, MMP-9 and MDR1 in peripheral blood to evaluate the value of chemosensitivity on breast cancer in vitro. Methods Fluorescent quantitative RT-PCR was adopted to detect the expression of CK19, MMP-9 and MDR1 in peripheral blood before and after chemotherapy. Results in experimental group and control group,the mean values of CK19, MMP-9 and MDR1 are -5.2105/-3.9938, -5.3312/-2.6930, 0.6750/3.6140 respectively. Independent-samples t test showed significant deviation between experimental group and control group (P 〈 0.01). Conclusion Besides the principle of Evidence-based Medicine, take the results of chemosensitivity in vitro as a factor to choicing antineoplastic combined chemotherapy protocols can increase the value of CK19, MMP-9 in peripheral blood and decrease the value of MDR1 more significantly.
