摘要
目的运用血清药理学方法观察脑康宁含药血清对原代培养的大鼠皮层神经元低糖低氧损伤的保护作用,进一步探讨分析脑康宁对脑缺血的保护作用机制。方法体外培养皮层神经细胞,建立低糖低氧损伤模型,通过MTT比色,测定培养液中乳酸脱氢酶(LDH)活性、NO和谷胱甘肽过氧化物酶(GSH-Px)的含量,观察脑康宁对神经细胞的保护作用。结果中、高剂量组含药血清均不同程度提高皮层神经元的存活率,能显著降低低糖低氧损伤所致的神经细胞培养液中LDH、NO和升高GSH-Px的含量。结论脑康宁对LDH、NO及GSH-Px的作用可能是其具有脑缺血保护作用的机制之一。
Objective To study the protection effect of Naokangning in serum on the primary cultured cerebral cortical neurons deprived of oxygen and glucose of rats, furthermore, approach the protection mechanism of Naokangning for cerebral ischemia. Methods The injury model of cultured neurons was made by the administration of sodium dithionite and glucose-deprived Earle' s solution. The protection effect of Naokangning in vitro analyzed with MTT reduction assay was observed by determining the content of nitric oxide (NO) by Griess reagent and the activities of lactate dehydrogenase (LDH) and glutathione peroxidase (GSH-Px) by LDH and GSH-Px kits, respectively. Results The survival of cortical neuron was raised with different degree in the middle and high dose groups. The MTT value and the level of GSH-Px in injured neurons were increased, but the LDH activity and the level of NO were decreased. Conclusion The effects of Naokangning on LDH, NO and GSH-Px may be a protection mechanism for cerebral ischemia.
