摘要
目的通过观察超氧化物歧化酶(SOD)在吸烟大鼠气道上皮细胞表达的水平,探讨SOD在吸烟所致慢性阻塞性肺疾病(COPD)氧化/抗氧化失衡中的作用。方法自制大鼠实验性被动吸烟装置,建立吸烟引起慢性阻塞性肺疾病的动物模型,将30只大鼠随机分为不吸烟组、吸烟1个月组、3个月组,每组10只。采用免疫组织化学染色半定量检测各组支气管上皮细胞Cu-ZnSOD蛋白的表达。结果吸烟3个月组大鼠出现了肺气肿的病理改变;吸烟1个月组(7.5±1.6)、吸烟3个月组(10.4±2.2)大鼠支气管上皮细胞Cu-ZnSOD蛋白的表达水平均较不吸烟组(2.6±0.4)明显增高,差异有统计学意义(P<0.01),3个月组较1个月组表达增高,差异有统计学意义(P<0.01)。结论随着吸烟时间的延长,大鼠支气管上皮细胞中Cu-ZnSOD蛋白表达增高,提示SOD在吸烟所致COPD氧化/抗氧化失衡中发挥一定作用。
Objective To evaluate the role of Cu-ZnSOD in course of oxidant/antioxidant mechanism in chronic obstructive pulmonary disease (CDPD) by observing the expression of Cu-ZnSOD in airway epithelial cell from smoking rats. Methods Rat smoking device was made by ,self. Wistar rats ( n = 30) were randomly divided into non-smoking group, 1 month smoking group, and 3 month smoking group (each, n = 10). The expression of Cu-ZnSOD protein on airway epithelial cells in 3-group rats was measured by immunohistochemistry. Results There was typical pathologic change of COPD in 3-month smoking group. The expression of Cu-ZnSOD in bronchial epithelial cells of incressed in 1- month (7.5±1.6)and 3-month (10.4 ± 2.2) smoking groulas compared to non-srnokig group (2.6 ± 0.4) (P〈 0.01). The expression of Cu-ZnSOD in bronchial epithelial cells in 3-month smoking group ( 10.4 ±2.2) increased compared to 1 month group(7.5 ± 1.6) (P 〈 0.01 ). Conclusion Cu-ZnSOD is likely to play an important role in smoking-induced oxidant/antioxidant mechanism in CDPD.
