摘要
目的研究氢溴酸高乌甲素对大鼠慢性缩窄性损伤(CCI)模型神经病理性疼痛的镇痛作用。方法24只大鼠建立CCI模型,分为四组,于术后第7天腹腔分别注射不同剂量的氢溴酸高乌甲素2、4和6mg/kg及生理盐水,给药后进行辐射热痛阈值的测定,测后取大鼠脊髓、脑组织标本采用EMSA进行核因子-κB(NF-κB)的活性测定。结果术后大鼠结扎侧足的热痛阈值明显缩短,氢溴酸高乌甲素4mg/kg组和6mg/kg组与生理盐水组相比较热痛觉过敏反应明显缓解(P<0·01)。NF-κB在生理盐水组中表达最高,2mg/kg组表达最低。结论氢溴酸高乌甲素能明显缓解CCI大鼠的热痛觉过敏现象,达到镇痛效果,同时能使NF-κB的表达减少。
Objective To study the analgesic effect of lappaconitine in rats with chronic constriction injury (CCI). Methods Twenty-four SD rats were randomly divided into four groups with six rats each :contrast group,group L1 (lappaconitine 2 mg/kg), group L2 (lappaconitine 4 mg/kg) and group L3 (lappaconitine 6 mg/kg). Hyperalgesia was measured by Plantar Analgesia Meter (Hargreaves Test) before operation and on the 1st, 3rd, 7th postoperative day. Seven days after CCI surgery, different dose of lappaconitine were administered. Hargreaves test was used to accesses the analgesic effect at 30, 60, 90, 120 and 150 minute after the drug administration. The animals were sacrificed immediately after test, NF-κB expression in the spinal and brain samples were analyzed by EMSA. Results After CCI surgery,all rats developed spontaneous pain, allodynia, and hyperalgesia. Injection of 4 mg/kg and 6 mg/kg lappaeonitine blocked the established hyperalgesia in the injured hindpaw. The expression of NF-κB in the spinal and brain was reduced in the rats given lappaconitine. Conclusion Lappaconitine could allay the rat's sciatic nerve cuffinduced hyperalgesla,and reduce the expression of NF-κB. Lappaconitine is effective in treating neuropathie pain.
