基质细胞衍生因子1及其受体CXCR_4在自体外周血造血干细胞动员中的表达及临床意义

摘要 目的探讨基质细胞衍生因子1及其受体CXCR4(SDF-1/CXCR4)在自体外周血造血干细胞动员中的表达及意义。方法采用免疫荧光标记和流式细胞仪表型检测分析CD3+4细胞上CXCR4的表达;ELISA方法动态分析患者造血干细胞动员过程中血浆中SDF-1水平的变化。结果①采集物中CD3+4CXCR4+细胞较动员前外周血和骨髓中CD3+4CXCR4+细胞均显著减少,分别为:(22.4±10.92)%,(32.09±12.71)%,(59.07%±26.98)%,P(0.01;动员好的组CD3+4CX-CR4+细胞数(20.11±8.87)%较动员差的组CD3+4CXCR4+细胞数(27.79±12.71)低,但无统计学意义(P=1.21)②血浆SDF-1至采集干细胞时显著降低;动员好的组血浆SDF-1平均含量显著低于动员差的组(P(0.001)。结论在造血动员过程中,SDF-1/CXCR4信号的表达是逐渐下调的,血浆SDF-1含量显著降低者动员效果好。 Objective To investigate the level and significance of stromal cell derived factor-1α (SDF-1α) and its receptor CXCR4 in patients peripheral blood during mobilization for peripheral blood stem cells harvesting. Method FACS analysis was used to study the expression of CXCR4 on the surface of CD34^+ cells and ELISA assay were employed to monitor the levels of soluble SDF-1. Results The mean percentage of CD3~ cells expressing CXCR4 in the apheresis collections was decreased in the PBSC collections compared with premubilization values ranging from 32.09 ± 5.39% to 22.4 ± 5.92%. There was a singnicant decrease in plasma levels of SDF-1 during the mobilization process;furthermore, SDF-1 levels in the apheresis collections corroborated with the with mobilization outcome. Conclusion SDF-1α/CXCR4 plays an important role during the procession of mobilization.
