摘要
目的利用5/6肾切除方法建立慢性肾衰竭动物模型。以中药复方制剂“肾纤复元胶囊”进行治疗,观察并评价治疗效果,探讨该药可能的作用机制。方法将Wistar大鼠随机分为假手术组、肾衰组和治疗组,后两组以5/6肾切除方法建立肾衰竭模型。治疗组手术次日给予“肾纤复元胶囊”300mg·kg-1·d-1灌胃,分2次给药,疗程12周;肾衰组及假手术组同时给予等量生理盐水灌胃。分别于第4、8、12周处死各组部分鼠取肾组织作α-SMA和Ⅰ型胶原染色(免疫组化ABC法)。结果治疗组大鼠一般情况优于肾衰组;治疗组4、8、12周BUN、Scr、24h尿蛋白定量明显低于肾衰组(P<0.01);光镜下肾组织病理切片显示治疗组损害轻于肾衰组,其α-SMA及Ⅰ型胶原染色阳性面积亦低于肾衰组。结论“肾纤复元胶囊”对慢性肾衰竭大鼠有较好的治疗效果,可延缓肾衰竭大鼠肾纤维化进程,减轻肾脏功能损害,保护残余肾功能。其作用机制可能为减少肌成纤维细胞的活化与形成,继而使细胞外基质胶原生成减少从而达到抗肾间质纤维化作用。
Objective:To confirm the effect of "shenxian fuyuan capsule"(SXFYC) on α- SMA and the Collagen Ⅰ. Expression in 5/6 Nephrectomy Rat. Methods:The aninmal model with chronic renal failure was establishced by the method of 5/6 nephrectomy. The rats were divided into 3 groups: treated group, control group and pseudo- operation (normal group). Next day after operation, rats in treated group were intragastrically administratd SXFYC 300 ·mg·kg^-1·d^-1. To contrast, rats in normal group and control group were fed on sodium chloride twice a day. After 4, 8 and 12 weeks, some rats were sacrificed and renal tisssues were examined by immunohistochemical staining for α- SMA and the Collagen Ⅰ. Results: Rats in the control group, which reeeived 5/6 nephrectomy, were in bad condition, accompanied by increased blood Scr and BUN in 4 weeks after operation. The general condition in the treated group were better than that in the control group. The parameters of Set, BUN and 24 hours' proteinuria at different time point of 4, 8, 12 week after operation in the treated group were much lesss than those in the control group (P〈 0.01 ). The positive: staining area of α-SMA,Collagen Ⅰ in the treated group was less than those in the control group. Conclusion: "Shenxian fuyuan capsulc" could ameliorate renal pathological damage in 5/6 nephrectomized rats and protect renal function of the remnant kidneys. It could decrease myofibroblast (MFIB) activation, transdifferentiation and collage production in the model of 5/6 nephrectornized rats.
