摘要
目的在基因水平上探讨中药橄榄降脂胶囊的降脂作用机制,为中药新药开发提供科学依据。方法将48只雄性SD大鼠根据血总胆固醇水平随机分为:正常组、模型组、血脂康组,及橄榄降脂胶囊低、中、高剂量组,每组各8只。正常组普通饲料喂养,其余各组予高脂饲料,自由饮水。模型组灌胃生理盐水,血脂康组灌胃血脂康胶囊0.09g/kg,橄榄降脂胶囊低、中、高剂量组分别灌胃橄榄降脂胶囊0.675g/kg、1.35g/kg、2.7g/kg。1次/d,连续给药6周。采用酶法检测大鼠血清TC、TG;采用磷钨酸-镁沉淀法检测HDL-c、LDL-c,RT-PCR法测定高脂血症大鼠肝脏高密度脂蛋白受体基因表达。结果模型组大鼠血清胆固醇、三酰甘油、低密度脂蛋白胆固醇均显著高于正常组,高密度脂蛋白胆固醇显著低于正常组(P<0.05)。模型组大鼠肝SR-B1mRNA表达降低,与正常组比较P<0.05。经橄榄降脂胶囊治疗后肝SR-B1mRNA表达升高与模型组比较P<0.05,与正常组P>0.05。结论橄榄降脂胶囊可调节脂质代谢紊乱、治疗高脂血症,其作用机理可能与上调肝SR-B1mRNA表达水平有关。
Objective To study the regulatory mechanism of lipid metabolism disorders in the blood fat of hyperlipemia rat model with Olive Antihyperlipidcmia Capsule, and do systematic observation on the functions of this medicine on lipids and high density lipoprotein receptor in rat liver gene expression, and then to clarify the mechanism of action of this medicine on treating hyperlipemia. Methods Select SD rats as investigated subject. The hyperlipemia rat models were made with feeding high-fat forage and were randomly divided into six groups based on the total cholesterol level at the rats' fasting for 12 hours; group A, group B, group C, group D, group E and group F. Each group included eight rats. Group A was normal group, group B was model group, group C was Xue Zhi Kang group, and group D, E and F were. Olive Antihyperlipidemia Capsule groups with low, medium and high dosage respectively. Group A was fed normal forage, but other groups were fed high fat forage. All groups were allowed to drink water freely. Since the first day when the models were made, intragastric administration had been adopted. Group A was fed normal forage without intragastric administration; group B was received physiological saline 1ml/kg.d with intragastric administration; group C received Xue Zhi Kang Capsule 0.09/kg .d with intragastric administration; Group D, E and F received Olive Antihyperlipidemia Capsule 0.675g/kg.d,1.35g/kg.d,2.7g/kg.d with intragastric administration respectively; each administered solution was 3ml,which comprised water and the certain medicine mentioned above, and administered for six weeks continuously. Each group was weighed once a week. At the end of the sixth week, and after the rats' fasting for 12 hours, the rats was anesthetized with pentobarbital sodium (40mg/kg) through abdominal cavity, .then the samples in the research were collected and examined the level of TC and TG in the blood serum by using enzyme; examined HDL-c, LDL-c by PTA-Mg2^+ precipitation method; analyzed the changes of SR-B1 gene expression in rat's liver by RT-PCR. Results Olive Antihyperlipidemia Capsule can markedly lower the level of TC,TG, LDC C in the blood serum, raise the level HDL, enhance SR-BI gene expression in rats' liver and finally accomplish the purpose of reducing blood fat. The experiment shows this medicine has the remarkable effect on hyperlipidemia, Conclusion Olive Antihyperlipidemia Capsule has an applicable value on preventing the cause, enhancing SR-B1 gene expression in rats' liver and finally accomplishing the purpose of reducing blood fat and development of hyperlipemia and its complications.
