Interleukin-1βand interleukin-1 receptor antagonist gene polymorphisms in Hu'nan Han nationoality ankylosing spondylitis patients

目的研究白细胞介素(IL)-1RN、IL-1β基因多态性与强直性脊柱炎(AS)的相关性。方法自100例AS患者及92名健康对照者外周血中提取DNA,采用PCR-SSP方法检测IL-IRN基因第二内含子VNTR复等位基因及IL-1β基因+3953单核苷酸多态性。结果AS患者中IL-IRN*2等位基因频率显著高于健康对照者。结论AS与IL-1RN*2等位基因相关联。

Objective To evaluate the relationship of the polymorphism of interleukin-1 receptor antagonist （IL-1RN） gene, IL-1β gene in patients with ankylosing spondylitis （AS）. Methods DNA 100 AS patients and 92 healthy controls were examined with PCR in a case-controlled study. Polymorphic sites in gene of the IL-1 family were examined, including IL-1RN VNTR and the SNP at position＋3953 in the IL-1β gene. Results No significant difference was seen at the polymorphic alleles in the IL-1β gene, but there was a significant increased IL-1RN VNTR＊2 allelic frequency in AS patients compared with controls （8.33% vs 3.26%, OR=2.697, P=0.027）. Conclusion The polymorphism of IL-1RN gene is strongly associated with the development of AS.
