摘要
目的:体外研究不同剂量脂多糖(LPS,lipopolysaccharide)对过敏性哮喘患者细胞因子分泌的调节作用。方法:随机选取过敏性哮喘患者8例,取其外周血3份,其中两份分别加入10μg、1μg的LPS进行24 h培养,设为高、低剂量实验组;另一份不加LPS同样培养,为对照组。ELISA法检测各组IL-4、IL-5、IL-10、IL-12及IFN-含量。结果:3组间IFN-生成量差异无统计学意义。与其他两组相比,高剂量组的IL-4、IL-5的生成量显著降低(P<0.05),而IL-10、IL-12生成量及IFN-/IL-4比值显著升高(P<0.05);低剂量组与对照组比较,IL-5显著性升高(P<0.05)。高剂量组IL-12与IL-4及IL-5呈负相关(r=-0.822,P<0.05;r=-0.768,P<0.05),与IL-10呈正相关(r=0.841,P<0.05);IL-10与IL-4及IL-5呈负相关(r=-0.812,P<0.05;r=-0.754,P<0.05)。结论:LPS调节过敏性哮喘患者细胞因子分泌具有剂量依赖性,高剂量LPS能有效调节过敏性哮喘患者细胞因子的分泌,抑制Th2型优势免疫。
Objective: To confirm the endotoxin dose- dependence of regulating the production of the cytokines of the peripheral blood in the allergic asthma patients. Methods: The allergic asthma patients with the middle severity or above were selected randomly and their heparinized venous peripheral blood were collected. The blood were cultul-ed with the high (10μg) or low (1μg) dose of LPS for 24 h, and the control group without LPS treatment was set up. The cytokines were measured by the enzyme- linked immmunosorbent assay. Results: (1) The differences of the levels of IL- 4, IL- 5, IL - 10, IL- 12 and IFN - γ/IL- 4 among three groups were significant except IFN - γ. (2) The high dose group have higher levels of IL- 10, IL - 12, IFN- γ/IL- 4 and lower levels of IL-4, IL-5 than the low dose group or control group (P〈0.05); the level of IL-5 in the low dose group was higher than control group (P〈 0.05), but the level of IFN - γ did not differ between them. (3) In high dose group, the concenmarion of IL- 12 was negatively correlated to that of IL- 4 and IL- 5 ( r = - 0.822, P 〈 0. 051 r = - 0.768, P 〈 0.05), possitively corrdated to that of IL- 10 ( r = 0.841, P 〈 0.05) ; IL- 10 was negatively correlated to IL - 4 and IL- 5 ( r = - 0.812, P〈0.05; r = -0.754, P〈0.05). Conclusion: LPS can dose-dependent modulate the cytokines secretion of the allergic asthma patients in vitro. High dose LPS lead to the increase in the levels of IL- 10, IL- 12 and the decrease in that of IL- 4, IL- 5, and inhibit the Th2 dcaninant immunity. There data suggested a new therapy approach of targeting the specific cytokines or other inflammatory mediators based on the LPS immune regulating functions for allergic asthma patients.
