摘要
蛋白质组学研究流程中蛋白质的鉴定是关键的一环,利用质谱进行蛋白质一级结构鉴定最重要的一步就是对蛋白质进行裂解。近年来,蛋白质氨基酸组成分析取得最有意义的进展之一是用微波辐射能进行蛋白质样品的水解。微波加热的机理与传统的分解方法相比有明显的优势,微波能量的快速吸收导致完全水解的时间大大缩短,从过去的几个小时到现在的几分钟。对近几年出现的一些微波辅助蛋白质组成分析新技术进行了简要评述,如不同溶剂中的微波辅助酶催化反应及气相微波辐射细胞壁水解等。微波技术在蛋白质鉴定中的新应用将在生物工程及蛋白质组学研究中起到更加重要的作用。
Protein identification is the key for proteome research and an important step is the hydrolysis of protein for MS analysis.Recently, one of the most important developments of the technology is the use of microwave radiation for sample hydrolysis, The mechanism by which microwave heating occurs offers certain advantages to conventional methods, the instant uptake of the radiation energy results in reduction of the, overall hydrolysis time from many hours to a few minutes, Here the progress in the last few years on microwave-assisted analysis of the compesition of protein techniques such as microwave- assisted enzyme- catalyzed reactions in various solvent systems, microwave cell wall hydrolysis in vapor phase are reviewed, This new application of microwave technology to protein identification will be an important advancement in biotechnology and proteome research.
