摘要
目的探讨甘露醇对急性肺损伤(ALI)兔循环内皮细胞(CEC)、IL-8表达的影响及其在ALI的防治作用和可能机制。方法以内毒素(LPS)致急性肺损伤兔为模型,随机分为3组对照组、甘露醇治疗组和速尿治疗组,分别观察4个不同时限点各组中肺湿重与干重比(W/D)、CEC及IL-8含量变化。结果甘露醇组肺湿重与干重比较对照组(即LPS组)显著降低(P<0.05),速尿组较对照组差异无显著性(P>0.05)。甘露醇组、速尿组各时限点CEC计数均低于对照组,6h时差异有显著性(P<0.05)。未经治疗的对照组,6h时CEC显著高于其他时限点(P<0.05)。IL-8检测结果显示,2h后对照组IL-8含量开始逐渐升高,并显著高于甘露醇组,与速尿组对比,差别无显著性。结论甘露醇可能通过影响炎症损伤级联反应的多个环节,抑制CEC、IL-8等多种效应因子的产生和表达,从而对ALI发挥防治作用。
[Objective] To investigate the effects of Mannitol on expression of CEC and IL-8 in the rabbits with ALI and explore the preventive-therapeutic effects of Mannitol and its possible mechanism. [Methods] Lipopolysaccharide (LPS) was injected into the rabbits to perform ALI animal model. The animals were randomly divided into three groups: the control group, the Mannitol treatment group and the Beronald treatment group. Lung wet/dry weight (W/D), the number of CEC and concentration of IL-8 in different group was respectively observed. [Results] W/D in Mannitol group was lower significantly than control group (LPS group) (P 〈0.05), while there was no significant difference between Beronald group and control group (P 〉0.05). In any time point, the count of CEC in both Mannitol group and Beronald group were lower than control group, especially in 6 h which had a significant difference (P 〈 0.05). In control group without cure, the count of CEC in 6 h was significantly higher than any other time point (P 〈 0.05). After 2 hours, the lever of IL-8 in control group had gradually increased which was significantly higher than Mannitol group, but compared with Beronald group there was no significant difference. [Conclusion] Mannitol has a protective effect on rabbits with ALI through effecting various cascade reaction of inflammatory injury and down regulating of the generation and expression of CEC and IL-8.
