藤茶总黄酮对小鼠的体内外抗氧化作用研究 被引量:39

摘要 目的:研究藤茶总黄酮(AGTF)的体内外抗氧化作用。方法:使用丙二醛(MDA)测定试剂盒测定AGTF对体内外MDA含量的影响;用分光光度法测定AGTF对H2O2诱导小鼠红细胞溶血和Fe^2+-VitC诱导的肝线粒体肿胀度的抑制作用。结果:2.00mg·mL^-1 AGTF对脂肪氧合酶的粗酶活性抑制率超过50%;AGTF在0.5~10.0mg·mL^-1范围内,具有较强的体外抗活性氧能力,并能减少小鼠肝线粒体及肝匀浆MDA的生成,可抑制H2O2诱导的小鼠红细胞溶血和Fe^2+-Vit C诱导的肝线粒体肿胀;腹腔注射AGTF对小鼠体内肝组织MDA生成的抑制作用显著。结论:AGTF具有较强的体内外抗氧化活性。 OBJECTIVE To evaluate the anti-oxidative effects of total flavonoids from Ampetopsis grosedentata (AGTF) in vivo and in vitro. METHODS The K3Fe(CN)6] model and the lipoxidase system were used to evaluate the deoxidizing capacity of AGTF and its inhibiting effect on lipoxidase activity. The effects of AGTF on the formation of MDA (malondialdehyde) and ROS(reactive oxygen species) in vivo and in vitro were detected by the reagent kits; and the hemolysis of mice RBC induced by H2O2 and the swelling of mice liver mitochondria induced by Fe^2+ -Vit C were determined by spectrophotometric methods. RESULTS The inhibiting rate of 2. 00 mg·mL^-1 AGTF on the crude lipoxidase activity was above 50%. 0. 5 - 10. 0mg,mLIAGTF could scavenge ROS in vitro, decrease the formation of MDA in mice liver and mitochondria, and inhibit the hemolysis of mice RBC and the swelling of mice liver mitochondria. In vivo, AGTF could significantly decrease MDA forma tion of mice liver homogenate. CONCLUSION AGTF has high anti-oxidative activity in vivo and in vitro.
