摘要
目的建立测定黄连中盐酸小檗碱含量的反相高效液相色谱法(RP-HPLC法)。方法采用HypersilC18柱(250mm×4.6mm,5μm),以磷酸盐缓冲液[0.05mol/L磷酸二氢钾和0.025mol/L十二烷基磺酸钠(1∶1),含0.2%三乙胺,并用磷酸调节pH值至3.0]-乙腈(60∶40)为流动相,流速为1.0mL/min,检测波长为264nm。结果盐酸小檗碱进样量线性范围是0.08936~0.53616μg,r=0.9998(n=6),平均回收率为98.6%,RSD为1.0%(n=6)。结论RP-HPLC法测定黄连中盐酸小檗碱含量,操作简单,结果准确,方法重现性好。
Objective To establish a HPLC method for determination of berberine hydrochloride in Coptis chinensis Franch,. Methods Hypersil Cra column (250 mm ×4.6 ram, 5 μm) was used; mobile phase was phosphate buffer solution [0. 05 mol/L KH2PO4 and 0.025 mol/L lauryl sulfonic acid sodium (l:l),containing 0. 2% triethylamine and adjusting pH value with phosphoric acid to 3.01- acetottrie(60: 40) ;detection wavelength was 264 nm;flow rate was 1.0 mL/min. Results There was good linearity in the range of 0,089 36-0. 536 16 tzg of berberine concentration (r=0.9998, n=6).Average recovery rate was 98.6%(RSD=l.0%,n=6),Conelusion The method is simple,accurate and good in repeatability.
