摘要
为探讨甘露醇的心肌保护作用机理,我们利用离体兔心非工作心脏模型,按改良的Langendorff法建立灌注装置,缺血停跳120分钟,再灌注45分钟。实验组采用标准ST·ThomasⅡ心麻液加甘露醇(36mmol/L),对照组加氯化钠(18mmol/L)以心肌含水量,(MDA)含量,心肌释放酶活性及超微结构为指标。结果显示:甘露醇组心肌释放酶活性及MDA含量明显降低,心肌含水量两组无明显差别,实验组超微结构明显优于对照组,表明甘露醇有明显的心肌保护作用,其机理主要不是渗透因素,而可能是自由基清除作用。
To elucidate the beneficial effect and mechanisam of mannitol,isolated rabbit heart non-working preparation by modified Langendorff model were subjected to 120′of hypothermic(17℃)cardioplegic arrest and 45′of normothermic(37℃)reperfusion.The expermimental group received standard ST. Thomas Ⅱcardioplegic solution with the addition of mannitol(36mmol/L),and in the control group by adding(18mmol/L).Myocardial edema.MDA, release of enzymes and electron microscopic changes were determined.Results show that mannitol treated hearts had less release of myocardial enzymes and production of MDA,but there is no marked difference in water content.The expermental group has better preservation of myocardial ultrastrutures than that of the control group. These results show the protective effect of mannitol may in part secondary to the free radical scavenging effect.
