结核分枝杆菌超声裂解产物体外诱导破骨细胞形成的相关实验研究 被引量:3

摘要 目的研究结核分枝杆菌超声裂解产物(Mt sonicate)体外诱导破骨细胞形成及其相关机制。方法通过破骨细胞计数、骨磨片吸收陷窝观察和TNF-α检测等方法观察不同浓度Mt soni- cate体外诱导破骨细胞的形成。结果Mt sonicate能够在体外诱导破骨细胞生长,且破骨细胞数量对Mt sonicate存在浓度依赖性。Mt sonicate诱导破骨细胞的形成机制可能与TNF-α的分泌增加有关。结论骨、关节结核的发病很可能与结核分枝杆菌诱导的破骨细胞增生有关。 Objective Tuberculosis of osteoarticular changes are often encountered in clinical work. Differentiation of osteoclasts may contribute to the pathological changes of the bone. The differentiation of osteoclasts were induced with Mycobacterial (Mt) sonicate to study the underlying mechanism. Methods Mycobacterial (Mt) sonicate was obtained from mycobacterial H37rv by sonicate clearage. Osteoclast counting,bone absorption pit measurement and TNF-α detection were used to evaluate the differentiation of osteoclasts. Various concentrations of Mt sonicate was used for induction. Results Mt sonicate induced the differentiation and proliferation of osteoclasts and Mt sonicates in a dose-dependent fashion. The mechanism of osteoclasts differentiation induced by Mt sonicate may involve induction of TNF-α. Conclusions The pathogensis of osteoarticular tuberculosis may involve osteoclasts proliferation and bone resorption induced by TNF-α secreting mycobacterium.
