摘要
目的研究外源性突变型p27(p27mt)基因对大肠癌体内抑制作用。方法以腺病毒为载体,采用细菌内同源重组方法制备Ad-p27mt,应用细胞移植法把人大肠癌SW480细胞接种于BALB/c裸鼠皮下,构建荷大肠癌裸鼠模型,将已成功构建的Ad-p27mt及Lac-Z采用瘤体内直接注射法注射到瘤体内,绘制肿瘤生长曲线,计算肿瘤生长抑制率,进行细胞凋亡及MMP-9检测。结果Ad-p27mt基因治疗组肿瘤生长明显受到抑制,有更强促凋亡作用,MMP-9表达明显降低,与Lac-Z、对照组比较,有显著性差异。结论Ad-p27mt基因对大肠癌肿瘤生长有明显抑制作用并能减少其转移。
Objective To study the inhibitory action of exogenous mutant p27 on colorectal carcinoma. Methods We constructed the recombinant adenovirus of human mutant p27 by the method of homogenous recombination in bacteria. The model of colorectal carcinoma nude mice was established by inoculating the SW 480 cells of human colorectal carcinoma to the subcutaneous part of BALB/c nude mice. The Ad-p27mt and Lac-Z were directly injected into the tumor of treatment groups respectively. The growth curves and the growth inhibition rate of tumor were analyzed. The cell apotosis and the expression of MMP-9 were detected. Results The Ad-p27mt gene therapy group.obviously inhibited the growth of tumor and promoted apotosis of SW 480 cells and degraded the expression of MMP-9 ,which were significantly different from those in the control group and Lac-z group ( P 〈 0. 01 ). Conclusion The Ad-p27mt gene can obviously inhibit the tumor growth and metastasis in the colorectal carcinoma in nude mice in vivo.
